Context of Genesis 48:6
4He said to me, ‘I am going to make you fruitful and will multiply you. I will make you into a group of nations, and I will give this land to your descendants as an everlasting possession.’ 5“Now, as for your two sons, who were born to you in the land of Egypt before I came to you in Egypt, they will be mine. Ephraim and Manasseh will be mine just as Reuben and Simeon are. Any children that you father after them will be yours; they will be listed under the names of their brothers in their inheritance. 7But as for me, when I was returning from Paddan, Rachel died – to my sorrow – in the land of Canaan. It happened along the way, some distance from Ephrath. So I buried her there on the way to Ephrath” (that is, Bethlehem). 8When Israel saw Joseph’s sons, he asked, “Who are these?”

The double portion in the inheritance is now transferred to Joseph. He is the first-born of her who was intended by Jacob to be his first and only wife. He has also been the means of saving all his father‘s house, even after he had been sold into slavery by his brethren. He has therefore, undeniable claims to this part of the first-born‘s rights. - Tyndale Open Study Notes · v. 5-7Jacob remembered Rachel (see 35:16–20) and blessed Joseph by making his two sons equal heirs with his other sons. The tribes of Ephraim and Manasseh would share equally with Jacob's other tribes (see Joshua 16–17). Jacob also gave Ephraim, Joseph’s younger son, the birthright (see 1 Chronicles 5:1–2).
